Seventy-nine UC-associated adenocarcinoma (UDCA) samples were compared with 79 sporadic adenocarcinoma (SDA) samples collected from surgical specimens. Samples were categorized into three groups according to colonic location (proximal, mid, and distal) and their stage of carcinogenesis (immature or mature). Immunohistochemistry was used to detect the expression of p53 and MDM2. p53 expression was only observed in 5% of immature UDCA samples and in 12% of mature UDCA samples. MDM2 expression was found in 10% of immature UDCA samples and in 11% of mature UDCA samples. MDM2 expression was significantly more frequent in UDCA than in SDA in proximal (P < 0.05) and distal (P < 0.05) locations and in immature UDCA than in mature UDCA in distal location (P < 0.05). MDM2 expression was more frequent in UDCA than in SDA in proximal location and in immature UDCA than in mature UDCA in mid location (P < 0.05). p53 expression was not related to age at diagnosis. p53 inactivation in early stages of UC-associated colon carcinogenesis does not seem to be the major mechanism of UDCA development.
