Chemical Substitution and the Nuclear Variant Phenotypes of the Wild-type and Mutant Forms of Oncoprotein BCL2 in B-cell Lymphomas. B-cell lymphomas are one of the most common forms of blood cancers. The study of B-cell lymphomas can provide useful insights into cancer biology, including both genetic and epigenetic components of the disease. A crucial factor in determining B-cell lymphoma aggressiveness is the balance between cell cycle regulation and apoptosis. Wild-type and mutated forms of the B-cell lymphoma-2 (BCL2) oncoprotein provide contrasting cellular phenotypes when overexpressed in cells. For instance, wild-type (wt) BCL2 promotes cell survival and inhibits cell death, whereas the BCL2 mutant (mt) BCL2 promotes cell death and inhibits cell survival. Unlike the BCL2 proteins, a number of other proteins are also known to play a role in the development of B-cell lymphomas. Here we present the results of a chemical substitution assay, which used metal-affinity chromatography, to establish the interaction between the wt and mt forms of BCL2 and a number of proteins known to be involved in B-cell lymphoma development. We found that multiple other proteins also interacted with wt BCL2, with the most notable interactors being the antiapoptotic proteins, BCL2 and BCL-XL. This is consistent with the fact that BCL2 and BCL-XL are mutated and/or overexpressed in many B-cell lymphomas. In the case of BCL2 mt, we discovered that only the small glycosylated protein associated with the mutant protein. This glycosylated protein is thought to regulate the kinetics of the BCL2 protein.
